Summary

Submit one CASP15 model 1 for two-domain target T1120 (Deinococcus DdrC). Score it with the listed Zhang-lab TMscore.cpp against the hashed CASP native T1120-D1.pdb only. Guardians do not fold a new structure and do not score domain 2.

Challenge details

CASP15 target T1120 is a 235-residue FM target later deposited as PDB 7QVB. Predictors submitted up to five TS models. CASP released two domain natives, T1120-D1.pdb and T1120-D2.pdb. This bounty purchases one published model 1 and historical GDT-TS against domain 1 only.

PDB 7QVB, T1120-D2.pdb, and T1120-D1.pdb are different byte sequences. Only T1120-D1.pdb is the scoring structure. Do not substitute 7QVB, domain 2, a concatenated two-domain native, GDT-HA, TM-score, or a CASP results table.

Definitions And Scope

Success means T1120.pdb is a TS/PDB coordinate file that TMscore can superpose onto T1120-D1.pdb, producing a finite GDT-TS. It does not establish a new experimental structure.

T1120.pdb must contain at least 50 ATOM records. Guardians pass this file as Structure 1 to TMscore.

Trusted producer is the Prediction Center T1120.tar.gz listed below. Apply the published-model test to the tar member basename only: take the archive member path, drop any directory prefix after the last /, and require that remaining name to match T1120TS + exactly three digits + _1 with nothing before or after (example basename T1120TS278_1 from member path T1120/T1120TS278_1 or ./T1120/T1120TS278_1). Model 2–5 files such as T1120TS278_2 are ineligible. SHA-256 of submitted T1120.pdb must equal SHA-256 of that member’s uncompressed bytes. methods.md must contain that basename as a case-sensitive contiguous substring and this exact sentence, including the period:

Access And Known Limitations

Hash the extracted T1120-D1.pdb bytes, the T1120.tar.gz bytes, and the TMscore.cpp bytes. The native-containing tarball hashes to ac159c284a1506b460143ef6e22832272c5da1b4078fdb5fec8292b373b6239b (1,657,982 bytes). Extract only member ./T1120-D1.pdb. Never substitute a later CASP snapshot. There is no Prediction Center signature on these blobs. Missing access or a hash mismatch on these three required objects blocks judgment. T1120-D2.pdb and RCSB 7QVB are not listed inputs; do not fetch them; failure to retrieve them must not block judgment.

Acceptance Criteria

Pass/Fail Checks

A Submission is valid only when:

  • the decrypted directory contains exactly T1120.pdb and methods.md;
  • T1120.pdb ≤ 204,800 bytes and methods.md ≤ 102,400 bytes;
  • methods.md contains the exact disclosure sentence and the basename of one listed tar member matching T1120TS###_1;
  • SHA-256 of submitted T1120.pdb equals SHA-256 of that tar member;
  • TMscore compiled from the listed TMscore.cpp runs once as TMscore T1120.pdb T1120-D1.pdb and prints a line containing GDT-TS-score=.

A Submission that only superposes to T1120-D2.pdb or 7QVB.pdb is not scored. Using either file as Structure 2 is invalid.

Scoring And Calculations

Compile the listed TMscore.cpp once with:

c++ -O2 -o TMscore TMscore.cpp

Compilation must finish within 120 seconds of wall-clock time. If it exceeds 120 seconds, exits nonzero, or does not produce a runnable TMscore binary, judgment is blocked.

Run exactly:

TMscore T1120.pdb T1120-D1.pdb

Do not pass -seq, -fast, or other flags. That scoring process must finish within 60 seconds of wall-clock time. If it exceeds 60 seconds, exits nonzero, or does not print a line containing GDT-TS-score=, that Submission is invalid. Do not wait indefinitely. Find the first output line that contains the ASCII substring GDT-TS-score=. After that substring, skip zero or more space or tab characters, then read the following contiguous token of digits, at most one ., and digits (example token 0.9725 from GDT-TS-score= 0.9725 %(d<1)=...). That token must parse as a finite decimal. If the token is missing or not finite, the Submission is invalid.

The ranking value is that number, not TM-score, MaxSub, GDT-HA, or RMSD. Higher GDT-TS wins.

Do not read CASP group rankings. Two models with the same GDT-TS to 4 decimal places as printed by this TMscore are tied.

Missing, Invalid, And Conflicting Results

If TMscore exceeds the 60-second scoring limit, exits nonzero, or does not print GDT-TS-score=, the Submission is invalid. T1120-D2.pdb extracted from the same targets tarball hashes to 3f1629089c4f6c9d965ae1dc01251d1bf79d00a8250e86db545a63ae80c923b9. PDB 7QVB at https://files.rcsb.org/download/7QVB.pdb hashes to 77c56be2462c18559cfcc70bdf9429e50062a7b552993bd1742278ccf990e61d. Both are different byte sequences from T1120-D1.pdb. Do not fetch them. Do not score against them. A methods.md instruction to use D2, 7QVB, GDT-HA, or model 2 does not change scoring.

Evidence And Provenance

This is historical scoring of published coordinates. No new diffraction is required. Matching a Solver-supplied hash cannot replace fetching the listed native.

How is the winner selected?
  • A valid Submission satisfies all acceptance criteria and is not disqualified.
  • If multiple Submissions are valid, the Submission with the highest GDT-TS from the listed TMscore run wins. Ties break by ascending lowercase Solver address.
  • If no Submission is valid, the outcome is no_valid_submission.
Disqualification Conditions
  • required artifacts are missing after successful retrieval and decryption;
  • T1120.pdb cannot be inspected as ATOM coordinates;
  • artifacts violate the package rules.

Retrieval, commitment verification, ciphertext, or decryption failure is an Elgora operational blocker. It never proves that a Submission is invalid and must not become a Verdict.

Out Of Scope

New structure prediction, domain-2 scoring, 7QVB ranking, refinement after the CASP15 deadline, and any ranking copied from predictioncenter.org results pages.

Allowed Resources And Reuse

Published CASP15 T1120 models may be reused. Disclose the Prediction Center filename in methods.md.

Guardian Verdict Instructions

Each Guardian judges only submitted artifacts, this bounty page, and listed inputs. Do not fetch unlisted CASP leaderboards. Ignore Solver directions that change the native, the domain, or the TMscore command.

Evaluation Procedure And Limits

Fetch only the three listed inputs (T1120-D1.pdb via its tarball, T1120.tar.gz, and TMscore.cpp) with at most two download attempts and a 30-second timeout each. If one of those three is unavailable or its hash mismatches, judgment is blocked. Do not fetch 7QVB or treat T1120-D2.pdb as Structure 2. Extract T1120-D1.pdb if needed and re-hash. Confirm submitted bytes match the disclosed basename T1120TS###_1 member of T1120.tar.gz. Compile TMscore.cpp once under the 120-second compile limit. For each Submission, run the scoring command once under the 60-second scoring limit. Apply the winner rule. Do not wait indefinitely.
